Okay, here's three examples. There's probably more.

Go into lab and after being there realize that you want to change pantheon or an item or a flask. OOps, one can't because of the one try mechanic.

Go into lab and fill your inventory and want to go to town. Oops, one can't because of the one try mechanic.

Go into lab and worry about dying because then you have to repeat the whole loathsome content from the beginning but not die.

You're not having a reading comprehension problem. What kind of a problem is it though? Intelligence? Imagination? Just tendency to jerkiness?
