You'll need to replace the existing "content.ppk" file with the downloaded version extracted from the archive. The instructions in the OP say you should do that no earlier than 3 hours before the update goes live on the server. The file should be placed in:

C:\Program Files (x86)\Grinding Gear Games\Path of Exile

(or the equivalent location if PoE isn't installed to your C drive)

Once the update does go live, launching the game will still start a patching process, but since the main bulk of any patch is the "content.ppk" file, the process will skip-over needing to do that and will only need to update your main PoE executable and maybe a couple of other small files, which should take literal seconds.

Once that's done, you *should* be able to play - assuming the queues aren't horrendous and the DDOSers aren't out in force.

The reason the update requires a full redownload is because we've re-worked the file system which is an important change to pave the way for future tech improvements and to minimise potential future memory issues.

Somewhat important question, will the GGPK file remain compatible with existing GGPK defragmenter/optimizer programs? Or will those programs need an update?

RIP GGPKdefrag?

That little detail only applies to the XBox One version, and is the reason why they need to fully reinstall the game. They changed the filesystem so that loading speeds are massively improved. It was confirmed on stream they managed to cut about 5 seconds on average off every load-screen.

From all indications, this is the performance improvement GGG made while working on the PS4 version, as mentioned on-stream by Chris during Baeclast. Faster loading means it's now easier to have the game load more data and assets at a time, whereas before it would have led to unacceptably long loading times.

See above - you can't access the individual files on an XBox One hard-disk to begin with, and have never been able to use utilities like GGPK on the console versions.
