Let me know what you think about this type of challenge. I think the glass cannon portion of it will make it fun and interesting in some respects. Obviously some classes will be more difficult than others but that is part of the fun!

If there are any questions, please feel free to ask! I will answer them when I can, and update this original post accordingly.

I think you're needlessly crowding out true melee skills with this challenge -_- but otherwise it seems fair and fun.

What if true melee skills like double strike allowed you to take up to +50% worth of pure defense nodes, and pseudo melee skills like ground slam and EQ could take +30%. Anything that hits the edge of the screen or off-screens has to do as you suggest.

Regardless of how you feel about that, you could add in the always thrilling, optional, "use a random number generator to determine your 2 main skills and your class".

The true melee skills IS a bit of a problem considering the challenge.

I was okkay with the idea since there are 'pseudo melee' skills as you put it, i.e. Lightning Strike, Molten Strike, Frost Blades, Wild Strike which are actual 'melee' skills that are projectile but at the same time marauder only has access to one of those. However, Marau has access to most of the other similar like sunder/EQ/GS/Spectral throw.

Perhaps going true melee in this challenge is the ultimate version of it? haha

I wanted there to be somewhat of an option for skills to use just because it makes it a little more open, especially with how true melee is almost a non-option, but if people wanted to use RNG to determine what class/skills they use it would make for an added challenge for sure.

I also wanted to add a stipulation that you can only pick up items for your class as well, and then go as far as to create loot filters that will only display items that are allowed for you to use in the challenge, that way you were only seeing the stuff you could actually use/sell.
I have never created a loot filter before, so maybe it would be a good practice.

I will be streaming this challenge about 2 hours from now. I will be winging a build for the most part...

Does anyone know if the PoE wiki has an updated list of every base type of armour/gloves etc?
Trust your gut.
Is mana leech or regen offensive?

Is life leech or regen offensive? What if using BM?

Which keystones are offensive? BM? EB? Avatar of Fire? Ghost Reaver? Necromantic Aegis?

Are utility nodes (eg: curse effectiveness/skill duration/ power or frenzy charge duration nodes, etc...) offensive?

Is movement speed offensive?

Are totem or minion life nodes offensive? What about +1 totem/minion nodes?

Are life nodes offensive if you plan to use RF? Are block nodes offensive if using tempest shield? Endurance charge nodes if using discharge?

Can you use un-offensive travel nodes if they're the only way to get an offensive node?
Face it, all of your suggestions are worse than this idea:

No, it would be considered similar to max mana/life

"

No, same as mana leech/regen, even if using BM

"

In general, all Keystones would be allowed unless they are strictly a bonus to defense.
BM would be allowed
EB would be allowed
Avatar of Fire would definitely be allowed
Ghost Reaver would be allowed
Necromantic Aegis would be allowed

"
Are utility nodes (eg: curse effectiveness/skill duration/ power or frenzy charge duration nodes, etc...) offensive?

All of these would be allowed with the exception of Endurance charges
Additional Endurance charges would NOT be allowed

"
Is movement speed offensive?

I thought that I had mentioned this in the OP, however as it turns out, I did not.
Movement speed would be allowed, but it would have to follow the same rules as stat points in that they are fine by themselves, but not when paired with a non-offensive/neutral trait

"
Are totem or minion life nodes offensive? What about +1 totem/minion nodes?

I am torn about this, because while they are defensive bonuses, if not allowed to use them, it would most likely make totem/minion builds almost completely useless and reduce that as a viable option for this challenge
I think it is best to go with:
Minion/Totem life nodes are allowed since it is boosting the viability of those specific options being used as an offensive tool... this is open to discussion
+# Minion/Totem would certainly be allowed

"
Are life nodes offensive if you plan to use RF? Are block nodes offensive if using tempest shield? Endurance charge nodes if using discharge?

This is a really good question, I have never played RF, so I am not sure if it would break the challenge or not to allow max life/es to be used as offense in this situation.
I want to say that it would still not be allowed, but that would essentially eliminate this build as an option I would think
For the time being, I am going to say that all of these scenarios would still NOT be allowed

"
Can you use un-offensive travel nodes if they're the only way to get an offensive node?

I would need an example for this situation because I can't think of exactly what you mean.
For the sake of ignorance I will simply say that it would not be allowed, but subject to change.

These are all REALLY GREAT questions/situations I had not considered, I will add a portion of the original post listing these specific situations and weather or not they would be allowed, as well as adding a portion of the post listing the questionable scenarios such as RF/minion-totem life/res

It is apparent that there needs to be some stipulations about what is explicitly considered to be offensive / neutral / defensive by nature. This will be addressed in the OP so we can all discuss it.
Trust your gut.
I like the idea, I don't think I have the patience for it though. Have you thought about any additional restrictions so its not like a "make a newbie talent tree" challenge?
Prejudice is a burden that confuses the past, threatens the future, and renders the present inaccessible.
You need to restrict:

All defensive flasks (with just flasks alone you can reach 50% dmg reduction and more for armor and evasion). And as you wrote, no unique items including flasks.
Cannot use fortify and endurance charges at all.

I would unlock mana and mana leech nodes (cant pick MoM though) since mana means outputing damage.

This was actually going to be my only question, but I kept trying to find examples and ended up being unsure what counted - which lead to those other Qs coming up :)

Anyway, here are some examples:

There is an AoE damage node that you can only get to by taking the scion starting node which gives mana regen and +5 int.

The 'solidity' node gives increased damage when holding a shield, but only has defensive nodes on either side of it.

The 'rampart' cluster as one node that only gives a boost to fortify.

'Devotion' gives increased curse effect, but is blocked off by life nodes on either side.

A non-scion can't get to 'path of the warrior' without taking some defensive nodes.
